Whole Plasmid Sequencing
Determine an entire plasmid sequence including its backbone, inserts and any structural
variations through long-read technology, without the need for primers.
ACGT offers complete plasmid and large DNA fragment sequencing of PCR amplicons or dsDNA viral genomes using long-read technology from Oxford Nanopore (ONT). This approach generates full-length target sequences using long-read de novo assembly, providing clear structural insight and high confidence results.

Bioinformatics Analysis
ACGT uses the latest ONT analysis pipelines to generate reliable consensus sequences. Once analysis is complete, you will be notified by email and can access your results by logging into your account.
Deliverables Include:
- FASTA consensus sequence files
- Interactive HTML sequencing report with plasmid map showing sequence features
Turnaround Time
Turnaround times are dependent on sample receipt at the Wheeling, IL laboratory by 10:00 am (CST) and same day pickup/delivery to the Germantown, MD laboratory. Samples received after the day's run time will be placed on the following day's run. We strongly recommend shipping samples via an ACGT drop box or through FedEx to ensure timely delivery. Please note that orders left in ACGT drop boxes that ship to Wheeling will be sent overnight for delivery the following morning.
| Turnaround Time | |
|---|---|
Monday - Friday Runs | Initial report will be made available by end-of-day on the run day* |
Troubleshooting | Samples that fail initial assembly will be re-tested one time free of charge on the next day’s run |
- | Raw data can be provided for download upon request |
*Turnaround times may be affected by events beyond ACGT’s control, including shipping delays and inclement weather. Samples received after 10:00 am on Friday will be run next Monday. Turnaround times may also be longer around ACGT-observed holidays. Customers can view holiday schedule changes in advance here.

WPS Free Trial
Test our data quality, turnaround time, and customer service with an offer of 2 FREE Whole Plasmid Sequencing reactions.*
* For first-time customers only. This offer is only valid for Standard WPS protocols with target sizes 25 kb or under; alternate workflows are excluded.
WPS Repeat Policy
If a sample fails to generate a successful assembly, it will be re-tested on the next run, using our troubleshooting workflow. If sample assembly still fails, or if the assembled sequence does not meet quality expectations, please contact our customer care team at [email protected]. Each request is reviewed individually to determine what corrective actions are available.
Additional quality assessments or corrective actions may include:
- Verification of DNA concentration, purity, and size*
- Magnetic bead–based sample cleanup*
- Sample analysis using alternative sequencing technology (Sanger or Illumina NGS)*
*Additional fees may apply
ACGT is committed to working with you to achieve successful sequencing outcomes.
